Comparación de métodos de extracción de ADN para el género Astylus (Coleoptera: Melyridae)

A successful extraction of DNA is fundamental for the progress of biologi- cal studies that include molecular identification, phylogenetic inference and genomics as well as sequences analyses. Although the methods of extraction of nucleic acids are universal, they must be adjusted according to the characteristics of each taxon studied. The aim of the present work is to compare DNA extraction methods and guide in the choice of the most convenient protocol according to the capacities of each laboratory. Four methods of DNA extraction were evaluated in the genus Astylus atromaculatus (Coleoptera: Melyridae) Taking as indicators DNA final concentration and quality, amount of starting material and, cost and time required. The evaluated methods were: 1) Commercial “HIGH Pure PCR Template Prepa- ration Kit” © Roche; 2) Saline precipitation method with SDS; 3) Saline precipitation method with CTAB and 4) Spin columns method. The four methods showed positive results regarding to the quantity and quality of DNA; with respect to starting material, the best result was provided by half abdomen and one segment of the leg. CTAB method was the best accord- ing to the concentration and quality of the DNA. SDS and SPIN COLUMNS methods showed intermediate values of concentration and purity, standing out the SPIN COLUMNS for its low cost. KIT extraction method showed the lower performance but the faster method.
